Abstract

The biochemical basis of defective step of cholesterol biosynthetic pathway for the cholesterol-dependent growth phenotype of the mouse myeloma, cell line has been studied.
In this studies, a cholesterol-dependent growth phenotype and a cholesterol independent growth variant of. mouse myeloma cells were labelled with [2-^14C] acetate and the distributions of radioactivity between cholesterol and its precursors were determined by thin layer chromatography.
The major labelled sterol product in the mouse myeloma cells after a 24 hr exposure to[2-14C] acetate was lanosterol and the major labelled sterol product in the variant cells after a 24 hr exposure to [2-14 C] acetate was cholesterol.
These results indicate that mouse myeloma cells are defective in cholesterol biosynthetic pathway and the site of defective is the step of.demethylation of lanosterol to C-29 sterol internediates.
